Investigating T cell populations for immune cell exhaustion in myalgic encephalomyelitis/chronic fatigue syndrome

Abstract Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is a devastating disease that confines ~65 million people worldwide to life of pain, severe exhaustion, cognitive deficits, flu-like symptoms, and postexertional malaise. We previously discovered decreased glycolysis, mitochondrial membrane potential, increased fatty acid oxidation in ME/CFS T cells. These features are commonly found exhausted cells, reversible state seen chronic viral infections decreases cell proliferation, survival, cytotoxicity. Our study evaluated whether subsets circulate with by analyzing CD4+ CD8+ cells for associated inhibitory receptors transcription factors. were isolated from 20 patients sedentary control blood samples measured ~15–17 markers at rest after stimulation using FACSymphony analyzer. Markers included population identifiers naïve, effector, memory, regulatory, helper significantly higher abundances frequencies factors related exhaustion general within PD-1+ samples. Half the cohort showed PD-1 memory than group. Moreover, exhibited levels Lag3, CTLA4, T-bet various subsets. This research detects novel immune dysfunction may suggest new treatment options.
